I'm assuming you're talking about when you press F8 or whatever the button is and you can go through the song without failing or pressing any arrows (the game does it for you, hence AUTOplay).
The only reason I have ever and could ever think about using autoplay is to get a feel for a new song. You could turn on the assist tick with the autoplay to get a good idea of the rhythm of the song and stuff, and then after a few times if you feel comfortable, you might play it.
But yea I think you just push F8 in-game and it will turn on.

ah, I knew it was F8, I just didn't know it was in-game. Another use is to put a billion retarded mods on a song and watch the computer pass it. Or Hellscaper for Nima

Hah, I do that sometimes. What I noticed is that on Autoplay the computer will pass it 100% with all marvs.
Put it on Jukebox mode with random mods on only challenge level songs. That's fun to watch.
